Determining Selectivity of PahZ1KT-1 in Polyaspartic Acid Hydrolysis

Dr. Bill R. Miller, Faculty Mentor

Poly Aspartic Acid Hydrolase 1/2 (PAAH1/2) is a dimeric protein that was discovered in river water bacteria, Sphingomonas sp. KT-1 (PahZ1KT-1), which has the ability to break down Poly Aspartic Acids (PAA) into their monomeric form. PAAH1 breaks up the PAAs from their long chain form to a smaller form. PAAH1 is broken down before PAAH2 is able to break it down further into its monomeric form. PAAs are bad for the environment because they are plastics that are water soluble, meaning they contaminate the water supply they are in. The goal of this project is to computationally simulate PAAH1 and PAAH2 catalytic domains and dock different length PAAs to these structures to find a good range for their binding affinity.
