Identification of In-Vivo Non-Telomeric Binding Sites for POT1

Human telomeres are composed of double-stranded (TTAGGG)n repeats and are capped by a single-stranded 3-telomeric overhang. POT1 (Protection of Telomeres-1) is known to bind the 3-overhang but not the double-stranded telomeric DNA. POT1 plays vital roles in protecting the 3-overhang and managing telomere length. POT1 contains two highly conserved oligonucleotide/oligosaccharide binding folds (OB folds) that bind to the 3-terminus of the G-rich telomeric overhang. Using SELEX, our group recently identified a non-telomeric ssDNA sequence motif for POT1 binding. The focus of this project is to determine if POT1 binds to the alternative DNA sequence motif in vivo. Using a technique known as ChIP-on-chip, we are working to identify the non-telomeric binding sites of POT1 in the human genome.
