Determination of the Ratio of Oxidized Cysteine to G-S

 ?-S is a protein that aggregates to form cataracts in the eye lens. Cystine, an oxidizing agent, could potentially help undo the aggregation process due to the extra cysteines located on ?-S.  Potential experiments include incubating DBF (disulfide bond-forming enzyme) and cystine with ?-S to see how effective cystine is at oxidizing DBF, which acts as a molecular chaperone to mediate protein refolding. 

Current experimentation includes incubating various ratios of ?-S and cystine to determine the ratio of ?-S and cystine necessary to facilitate the refolding process. ?-S and some buffer were used as the control and also incubated along with the other samples. The projected results are that a 5 to 1 ratio or less of cystine to ?-S will be able to appear on gels and will be enough to oxidize ?-S based on previous experiments.
