Enhancing Genome Editing in Eukaryotes

Cas9 mediated genome editing (CMGE) functions by targeting a specific region of the DNA and cutting it, thereby inducing repair with an artificial DNA molecule that contains a desired change. CMGE has been used to engineer new industrial strains of microbes and could eventually be used to treat genetic diseases. Unfortunately, success rates using CMGE are highly variable in eukaryotes. One factor that influences editing success is how tightly the target region of DNA is packaged onto nucleosomes. Herein, we investigate whether CMGE rates can be enhanced by nucleosome depletion using the drug trichostatin-A, a known global nucleosome depleter.
