Hydrophobic binding properties of Poloxamer-188 in electropermeabilized Rattus norvegicus flexor digitorum brevis myocytes

Extreme heat, electric shock and other methods of tissue-wounding have been known to result in cell damage and possible cell necrosis. Poloxamer 188 (P-188), a non-ionic triblock copolymer surfactant, has been widely used in the medical field as a lipid sealant for damaged cell membranes. Though P-188 binds and seals dismantled cell membranes, its specific binding mechanism remains uncertain. The objective of this study was to test the hydrophobic binding properties of P-188 by introducing the amphiphilic compound, lysophosphotydlcholine (lyso-PC), into an electropermeabilized rat (Rattus norvegicus) skeletal myocyte to inhibit the binding of P-188 to the exposed hydrophobic domain of the phospholipid bilayer. The results of the experiment suggest that lyso-PC does inhibit the binding of P-188 to the exposed damaged lipid bilayer.
