Transcriptome Analysis of the rld5409 Developmental Mutant of Maize

The maize rld5409 developmental mutant has narrow blades that are rolled upwards at the margins and epidermal abnormalities including: reduced numbers of macrohairs and buliform cells on the blade adaxial surface, abnormal subsidiary cells and stomata patterning, and regions of the abaxial surface that develop macrohairs. Our previous mapping studies demonstrated that the gene responsible of the rld5409 phenotype is rpb4. Rpb4 is a subunit of RNA Polymerase II, which is responsible for transcribing all protein coding genes. RNA-seq libraries were prepared from young leaf primorida that were captured from rld5409 and wild-type apices using laser microdissection. We used the Tuxedo pipeline to identify genes that are differentially regulated between mutant and wild type samples in order to identify genes that may also have a role in causing the observed rld5409 phenotype.
